Ziele/Aims The object of this module is to introduce students to independent scientific work in preparation for a potential master thesis. Students will work on their own small project under supervision of an experienced scientist and learn modern live cell and super-resolution imaging techniques as well as cell physiological experimental strategies. The focus of research in our lab lies on synaptic vesicle recycling and projects will be related to the field of synaptic vesicle exo-endocytosis.
Inhalte/Content During synaptic transmission small synaptic vesicles filled with neurotransmitter fuse with the plasma membrane to release their content. For maintaining synaptic transmission the exocytosed vesicle proteins have to be retrieved thereafter by compensatory endocytosis. Though intensively studied, many aspects of the exo-endocytic itinerary are still vividly debated. What is the fate of synaptic vesicle proteins post fusion? How are exo- and compensatory endocytosis coupled in time and space? Are there different vesicle populations supporting different modes of synaptic activity?
During this module, the participants will gain knowledge about the basic molecular mechanisms involved synaptic vesicle recycling and learn about experimental strategies to analyze these mechanisms in more detail. These strategies include e.g. application of different pH-sensitive fluorophores specific for SV proteins to monitor the dynamics during stimulation-dependent exo-endocytosis, and high-resolution fluorescence microscopy using photo-switchable dyes to analyze localization of distinct synaptic proteins with nanometer precision.
